NEXT WEBINAR: DIGITAL BIOREACTORS FOR FAST AND PREDICTABLE SCALING OF SHEAR-SENSITIVE CELL CULTURES

Written by: Mohi Ahmadinia

Introduction

Can you find the right operating conditions for your shear-sensitive cell culture before performing costly experiments?

In this webinar, we explore how Digital Bioreactors can enable faster and more predictable scale-up. The underlying technology uses advanced CFD simulations, providing a first-principles basis for predicting bioreactor performance. However, building a reliable Digital Bioreactor requires experimental validation. We will show how a range of experiments supported the development of a Digital Bioreactor of a 250 mL stirred-tank bioreactor. These included microcarrier suspension experiments, 4D particle tracking velocimetry (4D-PTV), and automated kLa measurements using Lucullus.

Once validated, the Digital Bioreactor serves as a virtual testbed that predicts how operating conditions affect suspension, mass transfer, and hydrodynamic stress. Rather than relying solely on trial-and-error experiments, we can screen operating conditions in-silico and identify promising experimental conditions, reducing the number of costly experiments needed during process development. Our principles can be applied to larger scales, supporting faster scale-up towards (clinical trial) production volumes. To demonstrate its practical value, we will show how the Digital Bioreactor guided the selection of operating conditions for iPSC-derived embryoid body formation, a process being developed for cRBC production.

What You Will Learn

During the webinar, Ramon will discuss:

  • Why conventional scale-up criteria fall short for shear-sensitive cultures
  • How a Digital Bioreactor is built and validated experimentally
  • How in-silico screening helps find optimal operating conditions across scales

Scaling up bioreactors for shear-sensitive cell cultures is time-consuming and costly. As bioreactor volume increases, achieving sufficient mixing and mass transfer without exceeding shear stress limits becomes increasingly difficult. This is particularly important for processes involving shear-sensitive cells growing on microcarriers or as aggregates (spheroids and embryoid bodies). Examples include processes using induced pluripotent stem cells (iPSCs; cell therapy), mesenchymal stem cells (MSCs, cell therapy), Vero cells (vaccines), and even bovine satellite cells (cultivated meat).
